Microscopy and Imaging Core Facility

Part 1: Confocal Microscope

Description:

Carl Zeiss LSM META 510 single photon confocal microscope. Excitation wavelengths produced by an Argon Laser (458, 477,488 and 514 nm), a HeNe-1 (543 nm), and a HeNe-2 laser (633 nm). Equipped with a 25x multimedia, 40x water, and 63x oil objectives. Accepts standard slides, 3 cm diameter dishes, and multi-well dishes on the stage.

Room 0031C on the Lower Ground Floor, Dental Sciences Building

Fee structure:

Training fee: $200 for a 4 hour session (mandatory)
User fee: $20 per hour

Part 2: Wide-field Optical and Fluorescence Imaging Systems

Description:

Three microscopy systems are available with complementary features, utilizing light sources and data acquisition systems from Photon Technology International.

• microscope-based fluorescence imaging station for monitoring regional changes of ions such as calcium, or other fluorescent markers
• dual excitation microscope-based photometry system for monitoring calcium concentration
• microscope-based imaging system for phase-contrast time-lapse recording of living cells.

System Features:

- Nikon Axiovert microscopes equipped with 10x, 20x, 40x, and 63x objectives;
- Micromanipulators for fine positioning of micropipettes; microscopes are mounted on anti-vibration air tables;
- Standard slides, 3 cm diameter dishes, and multi-well dishes can be placed in the stage;
- A stage warmer is available for live specimens contained in culture dishes; collections of single or time lapse fluorescence and phase-contrast images from live specimens;
- Eppendorf microinjector system is available for injecting proteins or probes into cells.

Training fee: $50 per hour
User fee: $5-20 per hour, depending on the system

Gene Expression Core Facility

Description:

Two instruments are available for analyses of gene expression. First, an Applied Biosystems Real Time PCR instrument, model 7900HT Sequence Detection System. This instrument is protected by a battery backup UPS. Second, a Tecan Safire microplate monochromator reader for absorption, and top and bottom fluorescence measurements.

Fee structure:

Training fee: $50 per hour
User fee for real time PCR: $50 per run
User fee for microplate reader: $5 per run
